The best routing from the various areas:

Joburg to Wartrail
If you are travelling by Car head down to Bloem and then take the N6 to Aliwal North and then R58 past Lady Grey and 44km after Lady Grey turn left to New England and follow the Lundeans Nek signs all the way to Wartrail. If you are coming down by Bike the best way is to find a route to Zastron on the R26 and head on the R393 and follow the Lundeans Nek signs.
 

KZN to Wartrail
By Car I would go to Kokstad and then head down the N2 and turn right to Tsolo on the R396 all the way to Maclear . Cars should turn left to Ugie on the R56 to Elliott and then on the R58 through Barkley east and just outside Barkey East turn right to wartrail and follow Lundeans Nek signs as well.If by Bike I would go over Naudes Nek to Rhodes  and turn right at Mosheshes Ford and onto Wartrail.

CT to Wartrail
Basically up the N1 and off to Graaf Rienet and through Hofmeyer , Molteno , Dordrecht to Barkley east and then before Barkley east turn left to Wartrail and Lundeans Nek  . I have not travelled this route but there are some Stop and Go's so check with AA.

PE to Wartrail
N2 to Grahamstown and then on the R63 to Fort Beaufort and onto Queenstown ,Lady Frere , Cala , Elliot and Barkley East . Go through Barkley and a few km out turn right to Wartrail and Lundeans Nek side.
